Acceso Abierto Association between GNRHR, LHR and IGF1 polymorphisms and timing of puberty in male Angus cattle(2012) Lirón, Juan Pedro; Prando, Alberto; Fernández, María Elena; Ripoli, María Verónica; Rogberg Muñoz, Andrés; Goszczynski, Daniel E.; Baldo, Andrés; Posik, Diego Manuel; Giovambattista, Guillermo; Peral García, PilarBackground: In bovines, there are significant differences within and among beef breeds in the time when bulls reach puberty. Although the timing of puberty is likely to be a multigenic trait, previous studies indicate that there may also be single genes that exert major effects on the timing of puberty within the general population. Despite its economic importance, there are not many SNPs or genetic markers associated with the age of puberty in male cattle. In the present work, we selected three candidate genes, GNRHR, LHR and IGF1, and associated their polymorphisms with the age of puberty in Angus male cattle.\nResults: After weaning, 276 Angus males were measured every month for weight (W), scrotal circumference (SC), sperm concentration (C) and percentage of motility (M). A total of 4 SNPs, two within GNRHR, one in LHR and one in IGF1 were genotyped using the pyrosequencing technique. IGF1-SnaBI SNP was significant associated (P < 0.01) with age at SC 28 cm, but it were not associated with age at M 10% and C 50 million. Genotype CC exhibited an average age at SC 28 cm of 7 and 11 days higher than CT (p = 0.037) and TT (p = 0.012), respectively. This SNP explained 1.5% of the genetic variance of age of puberty at SC28. LHR-I499L, GNRHR-SNP5 and GNRHR-SNP6 were not associated with any of the measurements. However, GNRHR haplotypes showed a suggestive association with age at SC 28 cm.\nConclusions: The findings presented here could support the hypothesis that IGF1 is a regulator of the arrival to puberty in male calves and is involved in the events that precede and initiate puberty in bull calves. Given that most studies in cattle, as well as in other mammals, were done in female, the present results are the first evidence of markers associated with age at puberty in male cattle. - Artículo
Acceso Abierto Cambios en propiedades químicas y bioquímicas del suelo asociados al reemplazo de pastizales naturales por Pinus radiata D. Don y rotaciones agrícolas(Universidad de Concepción (Chile), 2018) Di Gerónimo, Paula Florencia; Videla, Cecilia del Carmen; Fernández, María Elena; Zamuner, Ester Cristina; Laclau, PabloEl reemplazo de vegetación nativa de pastizales por agricultura, o por forestación, genera cambios en las propiedades del suelo afectando su productividad. El objetivo de este trabajo fue estudiar los cambios en algunas propiedades edáficas generados por los siguientes cambios en el uso de la tierra: 1) Pastizal Natural (PN) a agricultura (AGR); 2) PN a forestación con pinos (MP); y 3) Franjas raleadas dentro de MP: pastizal secundario (PS). Se tomaron muestras de suelo representativas de tales situaciones y se analizaron propiedades químicas y bioquímicas. Todos los sistemas produjeron una reducción del pH y aumento en la saturación con H+ del suelo. En los sitios MP y PS se produjo un aumento del carbono orgánico total del suelo (COT). El N potencialmente mineralizable por incubación anaeróbica (Nan) de estos sitios fue muy inferior al de PN, probablemente por diferencias en la calidad de su materia orgánica. PS disminuyó disminución en la acidez del suelo, y aumento del Nan y del P- extractable. En AGR se halló acidificación pero en menor grado, reducción del COT, Nan y P extractable. Los cambios observados indican que en sitios donde se reemplazó vegetación nativa por forestación con pinos, al retirarse la vegetación introducida, el suelo tiende rápidamente a recuperar sus características originales. - Documento de conferencia
Acceso Abierto Cambios en propiedades químicas y bioquímicas del suelo asociados al reemplazo de pastizales por forestaciones y agricultura(2016) Di Gerónimo, Paula; Videla, Cecilia; Laclau, Pablo; Fernández, María ElenaEn el Sudeste Bonaerense, el proceso de agriculturización implicó principalmente el reemplazo de vegetación nativa de pastizales por agricultura, mientras que, en los piedemontes serranos, algunos pastizales fueron reemplazados por forestaciones, debido a las restricciones que de estas áreas presentan para la agricultura. Otro cambio implementado, en algunos sitios forestados, fue la reconversión a sistemas silvopastoriles (SSP) mediante el raleo de franjas de árboles. El objetivo de este trabajo fue estudiar los cambios en algunas propiedades edáficas bajo las mismas condiciones ambientales y tipo de suelo similar, generados por los siguientes cambios en el uso de la tierra: 1) Pastizal Natural (PN) a agricultura (AGR) 2) PN a forestación con pinos (MP) y 3) Franjas raleadas dentro de MP, para reconversión a SSP: pastizal secundario (PS). Se tomaron muestras de suelo representativas de tales situaciones y se analizaron propiedades químicas y bioquímicas bioquímicas. Se observaron reducciones del pH y aumento en la saturación con H+ con respecto a PN, en todos los sistemas estudiados. En MP y PS se halló un aumento del carbono orgánico total del suelo (COT), principalmente de la fracción asociada a los minerales (COA), mientras que la fracción particulada (COP), no sufrió cambios con respecto a PN. El valor de Nan de estos sitios fue inferior al de PN, lo cual se atribuyó a ausencia de cambios en el COP y a diferencias en la calidad de los residuos. La apertura del monte (PS) generó disminución en la acidez del suelo y un aumento en la mineralización potencial de C y N, el Nan, y el P-Bray. En AGR se halló acidificación pero en menor grado, reducción del COT, principalmente del COP, reducción del Nan y aumento del P-Bray con respecto a PN. El estudio de las fracciones no logró explicar satisfactoriamente los cambios en COT, el Nan y la mineralización de C y N, en MP y PS. El estudio de la composición química de las fracciones ayudaría a su comprensión en futuras investigaciones. - Artículo
Acceso Abierto Comparison of the effectiveness of microsatellites and SNP panels for genetic identification, traceability and assessment of parentage in an inbred Angus herd(Sociedade Brasileira de Genética, 2013) Fernández, María Elena; Goszczynski, Daniel Estanislao; Lirón, Juan Pedro; Villegas Castagnasso, Egle Etel; Carino, Mónica Herminia; Ripoli, María Verónica; Rogberg Muñoz, Andrés; Posik, Diego Manuel; Peral García, Pilar; Giovambattista, GuillermoDuring the last decade, microsatellites (short tandem repeats or STRs) have been successfully used for animal genetic identification, traceability and paternity, although in recent year single nucleotide polymorphisms (SNPs) have been increasingly used for this purpose. An efficient SNP identification system requires a marker set with enough power to identify individuals and their parents. Genetic diagnostics generally include the analysis of related animals. In this work, the degree of information provided by SNPs for a consanguineous herd of cattle was compared with that provided by STRs. Thirty-six closely related Angus cattle were genotyped for 18 STRs and 116 SNPs. Cumulative SNPs exclusion power values (Q) for paternity and sample matching probability (MP) yielded values greater than 0.9998 and 4.32E-42, respectively. Generally 2-3 SNPs per STR were needed to obtain an equivalent Q value. The MP showed that 24 SNPs were equivalent to the ISAG (International Society for Animal Genetics) minimal recommended set of 12 STRs (MP ~ 10-11). These results provide valuable genetic data that support the consensus SNP panel for bovine genetic identification developed by the Parentage Recording Working Group of ICAR (International Committee for Animal Recording). - Artículo
Acceso Abierto La genética molecular de bovinos y equinos criollos en los albores del siglo XXI(Sociedad Argentina de Genética, 2010) Giovambattista, Guillermo; Rogberg-Muñoz, Andrés; Ripoli, María Verónica; Villegas-Castagnasso, Egle Etel; Díaz, Silvina; Posik, Diego Manuel; Lirón, Juan Pedro; Carino, Mónica Herminia; Goszczynski, Daniel E.; Fernández, María Elena; Peral García, PilarAmerican Creole bovines and equines are direct descendant from the animals introduced by European conquerors during XV an XVI centuries. At the beginning of the last decade, new technologies based in DNA emerged for genetic analysis, and Creole breeds were not the exception for these kind of studies. During the last years, these methodologies evolved vertiginously. In a short time, the amount of available information increased, and the analysis of few loci turned into analysis of complete genomes. This work describes the evolution of Creole, cattle and equines, molecular genetic studies during the last fifteen years. Even though, Creole breeds have not entered yet into genomic and proteomic era, its study is relevant because they are suffering a progressive population reduction. In this sense, information about infectious diseases resistance, adaptability, forensic genetics, animal production and phylogeography, among others, can be obtained from them. Furthermore, Creole breeds are a unique natural reservoir of genetic variability, and strategies for its conservation should be implemented.
